Physical Description
Ponceau 4R is a red dye that provides a dark red shade in applications. 
The color consists essentially of trisodium 2-hydroxy-1-(4-sulfonato-1- naphthylazo)-6,8-naphthalenedisulfonate, and subsidiary coloring matters together with sodium chloride and/or sodium sulfate as the principal uncolored components.  
Ponceau 4R is soluble in water and sparingly soluble in ethanol.

Typical applications include beverages, confectionery, bakery, dairy fats and oil, meat, seafood, snacks, dry mixes and seasonings, fruit preparation, convenient food, flavors and pharmaceuticals.

Ponceau 4r (e124) – Food Uses
Ponceau food color is synthesized using coal tar. 
Ponceau 4R is used to induce a red color in a variety of food products like sweets, jellies, desserts, tinned and canned fruits and foods, cakes, pastries, soups, soft drinks, canned beverages, etc.

Ponceau dyes are azo dyes meaning they are formed when two hydrocarbon groups are joined by two nitrogen atoms. 
The molecular formula for Ponceau 4R is C20H11N2Na3O10S3.
Ponceau 4R, like all azo dyes, is produced from petroleum products.

The chemical name of Ponceau 4R with the synonymous name Cochineal Red A is trisodium 2-hydroxy-1-(4-sulfonato-1-naphthylazo)-naphthalene-6,8-disulfonate with a structural formula as shown in Figure 2.1e. 
According to JECFA, the colour is manufactured by coupling diazotized naphthionic acid to G acid (2-naphthol-6,8-disulfonic acid) and converting the coupling product to the trisodium salt. 
Ponceau 4R may be converted to the corresponding aluminium lake under aqueous conditions by reacting aluminium oxide with the colouring matter.

As for the other azo-dyes, Ponceau 4R is authorised in a variety of foods including non-alcoholic flavoured drinks, with a maximum permitted level of 50 mg/l, and confectionary, fine bakery wares, edible ices, and mustard. 
The highest level permitted for the colouring of foods is authorised for decorations and coatings, sauces, salmon substitutes, and surimi with an amount of 500 mg/kg food, while edible cheese rinds and edible casing may contain the colour according to the quantum satis principle. 
Although typical use levels for many food additives are lower than the authorised maximum permitted levels, data from food industry indicate that Ponceau 4R is often used at levels close to the maximum permitted levels. 
Consequently, there are only small differences in exposure estimates based on maximum permitted levels compared to actual use levels. 
For adults, the typical exposure is in the range of 0.4–1.1 mg/kg bw/day (mean to high level exposure), while children have a higher exposure due to their lower body weight in the range of 0.3–6.7 mg/kg bw/day.

These agents are used to produce the desired color in the food articles. 
They include erythrosine, tartazine, quinoline yellow, sunset yellow, brilliant blue, indigo carmine, carmosine, Ponceau 4R, allura red, amaranth, etc. 
The allura red, quinoline yellow, and tartazine are linked to hypersensitivity reactions including asthma and skin rashes. 
In contrast, the erythrosine, sunset yellow, Ponceau red, indigo carmine, and brilliant blue have a potential role in carcinogenesis. 
Most of these agents are banned in many developed countries or their use has been restricted to fixed levels.

Ponceaue 4R interaction with protein, Nisin and BSA was concentration dependent and may be used for protein assay. 
As the dye binds with almost all the proteins and current methodology may be used for the estimation of proteins in various food systems. 
During the course of present work staining with ponceau 4R of resolved proteins on PAGE (poly acryl amide gel electrophorosis) was comparable with Coommassie Brilliant Blue R250. 
The Ponceaue 4R was highly sensitive, rapid and produced sharp red bands on the gel on 0.2% concentration. 
The effects of pH, concentration of proteins and dye were also investigated in various conditions which would help food processors to use a calculated amount of dye. 
The impact of tryptic digestibility on Ponceaue 4R -Protein Complexes (PPC) has illustrated that dye may safely be used without any adverse effect on the digestion of PPC.

Following a request from the European Commission, the Panel on Additives and Products or Substances used in Animal Feed (FEEDAP) was asked to deliver a scientific opinion on the safety and efficacy of ponceau 4R for cats, dogs and ornamental fish. 
The following ponceau 4R concentrations in complete feed were considered safe: 31 mg/kg for cats, 37 mg/kg for dogs and 137 mg/kg for ornamental fish. 
Inhalation exposure of ponceau 4R is regarded as hazardous. 
In the absence of data, the Panel cannot conclude on the irritancy potential of ponceau 4R to skin or eyes. 
No conclusion could be made on the skin sensitisation of ponceau 4R. 
Ponceau 4R is effective in adding colour to feedingstuffs.

Ponceau 4R is used as a coloring agent in many different products, such as food, drinks, medicines, cosmetics and tobacco. 
However, ponceau 4R also shows carcinogenic, teratogenic and mutagenic behavior in high doses. 
In this work, standard Raman, theoretical Raman and surface-enhanced Raman scattering (SERS) spectra have been used to investigate ponceau 4R. 
More specifically, density functional theory (DFT) calculations have been used to calculate the optimized Raman spectrum of ponceau 4R at the B3LYP/6-31G(d) level. 
This has provided a better understanding of the optimized geometry and vibrational frequencies of this dye. 

In addition, the experimental spectrum of ponceau 4R has been compared with the theoretical spectrum; good agreement was obtained. 
Finally, Ponceau 4R has shown that using SERS the detection limit of the ponceau 4R solution can be as low as 5 μg/mL. 
This has been achieved by SERS measurements of ponceau 4R on a substrate of gold nanoparticles. 
The SERS peaks at 1030, 1236, 1356 and 1502 cm(-1) were chosen as index for semi-quantitative analysis, showing that the SERS technique provided a useful ultrasensitive method for the detection of ponceau 4R.

Ponceau 4R is a synthetic colourant that may be used as a food colouring. 
Ponceau 4R is a strawberry red azo dye which can be used in a variety of food products, and is usually synthesized fromaromatic hydrocarbons; Ponceau 4R is stable to light, heat, and acid but fades in the presence of ascorbic acid.

Carmoisine (E 122), Ponceau 4R (E 124) and Allura red (E 129), are synthetic azo dyes 7generally used to give red colour to syrups, soda and sweets. 
They could be easily distinguished from the natural dyes, which are not electroactive, using differential pulse polarography. 
The influence of the pH on the intensities and the potentials of the peaks was studied between pH 3 and 11, and acidic or strongly basic media appeared not convenient. 
Ponceau 4R was shown that in a pH=9 phosphate buffer, the peaks of Carmoisine, Allura red and Ponceau 4R were well shaped and separated, allowing accurate identification and quantification, even if the three dyes were mixed. 
No significant changes of the peak potentials were noticed in the commercial samples, and consequently the dyes can be identified without ambiguity. 
A procedure using the standard addition technique was validated with test syrups.

The reddish-orange dye is known to cause cancer, allergic reactions and asthma. 
Presently Ponceau 4R is banned in Canada, Norway and United States. 
Ponceau 4R is an azo dye which is produced from petroleum products.
In our latest test we found five samples of dried prawns, two samples of fish crackers and one sample of char siew (barbecued pork) containing Ponceau 4R. 
The samples were bought in Penang, Kedah and Perlis. 
The char siew sample was bought in Penang.
In our earlier tests we found the dye to be used in turmeric and curry powder. 
The dye was also used to adulterate tea.
